The determination of pentamidine (4,4′-diamidinophenoxypentane) in plasma, urine, and tissues

Abstract

A sensitive fluorometric method for pentamidine assay in human plasma and urine and in mouse tissues and urine is described. Pentamidine is extracted under basic conditions from plasma, urine, and homogenized tissue samples into organic solvents. Following extraction and concentration into acid, the reaction between the pentamidine, glyoxal, and benzaldehyde to form a fluorescent glyoxalidone derivative is carried out in basic solution. After extraction of this product into organic solvents, the final determination is made with the use of a spectrophotofluorometer.
