Abstract
There has been no effective strategy for preventing restenosis in man. Therapeutic strategies have focused on limiting intimal hyperplasia and its physiologic consequences. Although many drugs have been tested with some success in animal models, none have been effective in preventing restenosis in clinical trials. In this study, two hydrophobic drugs, Galardin and Taxol, are evaluated for their effect on neointimal hyperoplasia after loading into gelatin-chondroitin sulfate films. Galardin is an experimental peptide analogue, which has recently been shown to inhibit matrix metalloproteinases; Taxol is an antineoplastic drug, which has been shown to inhibit neointimal proliferation in the rat when delivered systematically. Two animal models, rat and minipig, have been used to test the effects of Galardin and Taxol for restenosis.
