Strategy for the analysis of troponins in cardiac tissue using a combination of affinity chromatography and MS

Abstract

The isolation of the cardiac troponins (TnI) and its modified products, from diseased heart tissue using affinity chromatography (AC) and mass spectrometry, was discussed. The AC was performed using the anti-TnI antibody 81-7 coupled to NHS-activated Sepharose beads. The AC of the cardiac tissue homogenate faciltated the specific separation of TnI and proteins bound to it. It was found that the methods were able to identify not only the TnI, but also both of the other trophonin subunits, TnT and TnC.
