Multimodal thin layer chromatographic separation of bryostatins 1 and 2 from Bugula neritina extracts employing side-by-side silica gel/C18 plates

Abstract

The separation of bryostatin 1 from bryostatin 2 and other compounds from Bugula neritina extracts using silica gel/C18 side-by-side thin layer chromatography plates was accomplished. Clean-up and preliminary separation was carried out on the silica gel strip with a mobile phase of hexane:acetone 3:2, while resolution was achieved on the C18 side of the plate using 85% acetonitrile/water. Visualization of bryostatins 1 and 2 was achieved by spraying with anisaldehyde reagent and heating in an oven at 110°C.
