Linkage studies and prediction of risks for Huntington Disease

Abstract

Data from the first three pedigrees were analyzed for linkage between Huntington's Disease (HD) and the DNA marker, G8. The effects of different age-of-onset curves and different gene frequencies for the G8 haplotypes were studied. For each pedigree, two different age-of-onset curves for HD were used in the analyses. Also, the frequency for the 'affected' haplotype was both increased and decreased to assess the effects of different gene frequencies on the linkage results. In addition to the linkage analyses, a preliminary study was done to determine risk for HD in at-risk family members. To simulate a typical counseling situation where less extensive pedigrees will be available, the probability of developing HD based on a limited amount of family data was calculated for several young at-risk subjects in Family 3. Effects of different estimates of the recombination fraction, different age-of-onset curves and different frequencies for the G8 haplotypes on the probability of carrying the HD gene were investigated.
