Abstract
A rapid semimicro assay for lymphocyte activation in man is described. It is based upon the stimulation of protein synthesis in peripheral blood lymphocytes cultured in serum free medium with soluble antigens, phytomitogens or allogeneic lymphocytes. Cellular response to PHA is discernible in 4 hr and response to antigen in 16 to 24 hr. Stimulation in mixed lymphocyte cultures can be detected as early as 8 to 10 hr and invariably by 42 hr. The assay permits detection of lymphocyte activation earlier and with greater precision than the widely employed thymidine incorporation blast transformation system.
